Host File Access & Browser Hijack

I ran Spy Bot and it located problems with "Microsoft Windows Redirected Hosts" and "Fraud Windows Protection Suite."

However, I get an error report when trying to fix the problem --

Cannot create file c:windows\system32\drivers\etc\hosts.

I know the browsers are hijacked. I am close to reformatting but am also concerned about whether that will actually clear the problem.

If anyone can assist it would be most appreciated.
