Default Immunization after reboot needs redone

Windows 7 user with Spybot w/anti-virus version 2.4.40.0
Each time I open Spybot and run immunizations it checks my system and shows that I have a large number that are not immunized, so I have to reapply.
Is there something I need to do to have them stick? I empty my recent Items, History, and Cache before each shut down of the system.
Would running Malwarebytes and Microsoft Security with Spybot at the same time have a bad effect causing this?
I only use Firefox and empty the cookies and history also.

Are you running Spybot as an administrator?

You can do this by right-clicking the Spybot icon and choosing the option to "run as administrator". This must be done to give Spybot the permissions it needs to edit the hosts file to apply the immunization.
